What is 4-AcO-MET?
4-acetoxy-N-ethyl-N-methyltryptamine (also known as 4-AcO-MET, metacetin and azomet) is a lesser-known novel psychedelic substance from the tryptamine class. Members of this group produce psilocybin-like psychedelic effects when administered. It is structurally related to psychedelic tryptamines such as 4-AcO-DMT, 4-AcO-DET and 4-AcO-MiPT.
4-AcO-MET is structurally closely related to 4-HO-MET, for which it theoretically acts as a prodrug.
4-AcO-MET occasionally occurs in pressed pills that are sold on the street in northern Switzerland under the name “Acomet” or “Azomet”.
Very little data is available on the pharmacological properties, metabolism, and toxicity of 4-AcO-MET, and there is little history of human use. It is sold online as a research chemical. It is strongly recommended that harm reduction measures be used when using this substance.
Chemical properties
4-AcO-MET or 4-acetoxy-N-methyl-N-ethyltryptamine is a synthetic indole alkaloid molecule of the tryptamine class. Tryptamines share a common core structure consisting of a bicyclic indole heterocycle attached to an amino group at R3 through an ethyl side chain. 4-AcO-MET is substituted at R4 of its indole heterocycle with an acetoxy (AcO) CH3COO− functional group. It also contains a methyl group and an ethyl chain attached to the terminal amine RN of its tryptamine backbone (MET). 4-AcO-MET is an acetate ester analogue of 4-HO-MET and the N-substituted ethyl homologue of 4-AcO-DMT.
Pharmacological effects
The psychedelic effects of 4-AcO-MET are thought to be due to its potency as a partial agonist at the 5-HT2A receptor. However, the role of these interactions and how they lead to the psychedelic experience remains unclear.
It is also suspected that this compound is rapidly hydrolyzed to the free phenol 4-HO-MET, although human studies on the metabolic fate of this drug are lacking. This would explain a somewhat similar experience in its subjective effects. This is similar to the presumed deacetylation of 4-AcO-DMT to 4-HO-DMT during first-pass metabolism and subsequent passage through the liver.
